Spinal cord injury (SCI) results in lifelong disability with severe socioeconomic consequences and remains untreated to date. The complexity of SCI impedes repair through poor revascularization, release of cytotoxic molecules, and cavity formation; exacerbated by persistent ischemia and inflammation. Thus, effective therapies will need to combine synergistic approaches to overcome these multiple obstacles. In this thesis project, we propose to combine two strategies that each have proven beneficial effects to treat spinal cord injury: (i) implantation of a degradable natural hydrogel designed as a scaffold capable of inducing tissue remodeling and low intensity repetitive magnetic stimulation (LIrMS). One of our objectives is to directly determine the involvement of endogenous ependymal spinal stem cells (Endo-Ssc) and the effects of our strategy on their role in repair. This will be facilitated by the use of inducible transgenic mice to block their proliferation. In the rat SCI model, which is more similar to human SCI, we will evaluate the results by sensorimotor, advanced imaging and molecular analyses. The project, based on a strong collaboration between three distinct teams, is at the interface of neurobiology and materials science and will develop a state-of-the-art regenerative strategy for future clinical use.

The PhD student will be in charge of the realization of this project. He/she will have to perform the surgery on rats and mice, to set up the different treatments and to perform the functional evaluation of the animals over time. In addition, the student will perform histology and microscopy experiments as well as image processing. He/she will also be in charge of the analysis of the different results obtained, their writing and presentation.
